Resource genetics idendtification is one of the most efficient strategies for fisheries management regarding commercially important marine resources.One of the valuable species of shrimp in the Persian Gulf is the banana shrimp (Fenneropenaeus merguiensis), which accounts for about 60% of total annual shrimp catch in Hormozgan province.Identifying the genetic structure of the country's marine reserves is an effective way of efficient fisheries management. In this study, the amount of genetic diversity in the populations of this species was studied using microsatellite markers by 10 primers in four populations at Jask, Kolahi, Bandar Abbas power plant and Tula catch area and their genetic characteristics were compared with a 1 gram shrimp batch as a sample from stock enhancement project which was being stocked near kolahi area. Results of Genetic Distance Matrix, Heterozygosity, Assignment Test and Hardy-Weinberg Equilibrium at the Locations Indicated Average Population Distance (over 0.05 ) and Possible Effects of nursery ground and Regional Hydrodynamic Flows.

In this study, two populations of Aphanius ginaonis (Holly, 1929) and Aphanius dispar (Ruppell, 1828) were examined to determine genetic diversity using PCR-RFLP technique. A total of 60 individual specimens were collected from Hormozgan and Bushehr internal waters, comprising two sampling sites; from Geno hot spring (30 individuals of A. ginaonis) and Mir Ahmad hot spring (30 individuals of A. dispar). The D-loop region of mitochondrial DNA (550 bp) was amplified using PCR followed by RFLP analysis based on 5 restriction endonucleas enzymes (AluI, DpnI, Eco47I, HindIII HinfI). In order to undertake data analysis, Arlequin 3.11 was applied. Results at population levels indicated that Geno individuals have more haplotype diversity than Mir Ahmad individuals (9 and 3 haplotypes respectively). However, further investigation using genetic techniques is required to clarify the molecular history and evolution of Aphanius species in this area. Results obtained from this research would be applicable to understand conservation genetics and management of this important fish species in Iran.

Brown algae of Padina sp (Adanson, 1763) are found globally across tropical coastal areas, inter-tidal and sub-tidal regions. The aim of this research was to identify morphological and molecular characteristics of padina species distributed across Lengeh Port in Persian Gulf using morphological examinations and rbcL chloroplast gene sequencing. For this purpose, morphological features were undertaken using valid identification keys. For molecular analysis, total genomic DNA was extracted by CTAB method with slight modifications. Amplification of fragments was carried out using rbcL primers. Analysis of genome sequences was undertaken using genetic softwares of Chromas, BioEdit and MEGA6 and phylogenetic trees were constructed by two models of Neighbor Joining (NJ) and Maximum Likelihood (ML). Results indicated that two identical species of padina (P. boergesenii and P. australis) in this research shared common cluster with 99% probability and showed high genetic similarities among related compared padina species submitted in NCBI Genbank.

16S rRNA is one of the components of the ribosome and widely used in phylogenetic studies of closely related species and especially related species Crassostrea. Nucleotide sequence polymorphism associated with 16S rRNA gene of mitochondrial genome of Persian Gulf Oyster (Crassostrea sp.), examined in 30 samples collected from Badar-e Emam Khomeini oysters. Through a primer PCR chain reaction was performed for all samples and product of PCR were Automated sequencing. Then the data obtained were analyzed by the programs and software. For the gene studied only there was 2 mutation and Nucleotide diversity (Pi) 0.00061 calculated. Among the 30 samples, 3 haplotypes were detected and average haplotype diversity (Hd) 0.195 were calculated. The haplotypes obtained in this study were first registered in the gene bank. Phylogeny trees drived from 16S rRNA gene sequencing showed no clear separation of the samples in the Bandar-e Imam Khomeini region. The results of this study showed that there is low sequence differentiation in the study population in Bandar-e Imam Khomeini.

The purpose of this study was to evaluate the antioxidant properties of hydroalchoholic extractions of three species of green, brown and red algae. The highest and the lowest total phenol contents was in L. snyderia (113/9±0/69 mg gallic acid per gram of extract) and the green algae E. intestinalis (72/36±6/05 mg gallic acid per gram of extract) respectively. The highest values of total flavonoid was founded in the red algae L. snyderia (41/05±1/95 mg Rutin per gram of extract) however, the lowest values was (12/7±0/41 mg Rutin per gram of extract) in the brown alga C. trinodis. L. snyderia showed the most antioxidant activity, and C. trinodis had lowest amounts of antioxidant potential by the radical, azinobis ethylen benz thiazoline sulphonic acid (ABTS) test. there were significant differences between anti-oxidant activities of algae according the ABTS test.

The purpose of this study was to evaluate of bio diversity and density of brown algae in coast of Bushehr province in the Persian Gulf during warm and cold seasons. The most and lowest density of brown algae was in the spring (33±20Number in m2) and in autumn(17± 9Number in m2) respectively. The highest density was belonged to species of Cystoserria myrica with(41±25)and it was dominant species among the six species of brown algae were sampled. The most of Margalef index(0/438) was observe at the mid, and mid litoral zone. The maximum value of the Shannon index(0/94) was belonged to the low, mid litoral zone. Most of species richness and dominance was revealed at autumn and highest indices of Shannon(0/76) was at spring. Brown algae in the spring had the most density (33±20/86 Number in m2). Species richness was detected the highest and lowest in Nuclear power Plant and Ganaveh stations in two seasons respectively. Higher levels of dominance and low diversity was in within the tidal zone.

Caviar-producing fish with their economically valuable product are important in fisheries. The cDNA growth hormone (GH) of Sturgeon fish Huso huso was constructed using total RNA from pituitary glands. To construct the recombinant and active lentiviruses carring GH gene, this DNA sequence was inserted into the cloning vector pTZ57R/T and subsequently cutted from pTZ57R/T by endonuclease enzyme and incorporated into lentivirus vector pNL-EGFP/CMV-WPRE on upstream of an IRES cassette. We also insert a reporter EGFP gene downstream of IRES so transfection and transduction steps can be traced. Using this vector plus virus packaging and envelope vectors, HEK-293T cells was co-transfected by DNA-Lipofectamine complex method. Cell supernatant full of virions was collected 48 hours later and concentrated using Amicon columns to obtain a high-titer virus stock. Nearly 1/5 of this stock was applied to a new batch of cultured HEK-293T. After 72h expression of EGFP gene was Detected and the cells was collected for further analysis. Total RNA of these transduced cells was Extracted and GH mRNA expression was revealed by RT-PCR. Results showed that, lentiviral vectors (LV) as a gene transfer system provide efficient delivery, integration and long-term expression by establishing a stable provirus in target cells and could be An important tool in aquaculture and fisheries biotechnology research to increase the growth rate of farmed fish by transferring growth hormone (GH) transgenes into fish.

Total genomic DNA was extracted by proteinase-K digestion, phenol: chloroform: isoamyl alcohol extraction and ethanol precipitation. The quality and quantity of DNA was assessed using 1% agarose gel electrophoresis and spectrophotomeric method. Polymerase chain reactions (PCR) were conducted on the target DNA using 6 primers RAPD. PCR products were separated by electrophoresis on polyacrylamid gels (8%) that were stained using silver nitrate. Allele frequency, the effective and real allele, expected and observed heterozygosis, genetic diversity, Shannon Information index, Gst and gene based on analysis of molecular variance (AMOVA) were calculated using the GENALEX and Pop Gene software. The average of Shannon index for Hendijan, Khore zangi and Delvar was respectively 0.3887, 0.2860 and 0.3509. Highest of Gst observed between Hendijan and Khore zangi (0.260) and lowest between Khore zangi and Delvar (0.195). Highest of gene flow (Nm) was observed between Delvar and Khore zangi (1.033) and lowest between Hendijan and Khore zangi (0.687). Highest of Genetic identity observed between Delvar and Khore zangi (0.924) but lowest was between Hendijan and Khore zangi community (0.833). Eventually this study show that three community of our research are separate and belong to two clusters.

Heavy metals are major group of dangerous contaminants in marine environment. Due to their persistence, they may enter aquatic food chains and transfer from one trophic level to another. They also may be harmful for human being which is placed at the top of the food chain. Imam Khomeini’s Port is located in the north west end of Persian Gulf and ships traffic and several industries especially petrochemical industries around of the port, discharge various organic and non-organic contaminants such as heavy metals into this ecosystem. In this study in order to investigate the levels of heavy metals (Hg, Cd, Pb and Cu) in soft tissue of oyster Crassostrea gigas from Imam Khomeini port, oysters of the same size were sampled on March 2010 from 5 different locations including harbors of Petroshimi, Doc Sorsoreh, 15, 28 and 33 in the Imam Khomeini port. The samples were freeze dried and acid digested in order to determine their heavy metal content with atomic absorption spectrophotometry. Results indicated that the content of Hg, Cd, Pb and Cu in the oysters’ soft tissue were 3.12±2.97, 7.56±3.62, 7.1±3.30 and 440.57±66.17 µg/g of dry weight, respectively. The highest level of Hg, Pb and Cu were recorded in harbor of Petroshimi while the highest level of Cd was measured in harbor no 15.The concentration of Hg, Cd, Cu and Pb in bivalve soft tissue was more than standard limits. This can be dangerous for public health therefore regular monitoring of heavy metals in the area is recommended.

The Mendelian Inheritance of produced hybrids from Chalcalburnus chalcoides and Vimba vimba persa was investigated using Microsatellite Markers. Genomic DNA from the produced larva and also small piece of dorsal and pectoral fin of their parents were extracted by Phenol-Chlorophorm method and PCR reaction was accomplished. 10 loci were amplified, in which 7 of them were amplified with reasonable polymorphism and 3 were monomorphism.
In order to study the segregation of the produced F1 hybrids from female Chalcalburnus chalcoides and male Vimba vimba persa, seven loci (LCO3,LCO1,Lid11,Rru2,Z1-2,Z7-8,Z9-10) were used.
According to the results it showed that parents and F1 larva‘s allelic distribution were almost segregated based on Mendelian inheritance in 7 loci. The present study showed that Microsatellite markers are useful marker in study of inheritance.
